Everyday meaning is as follows: "Be rational" usually means “be reasonable” or “think logically.” "Get real" is a slangy way of saying “be realistic” or “stop being ridiculous.” In the cartoon: i says to π: “Be rational” — which is ironic because π is irrational in math. π replies: “Get real” — which is ironic because i is imaginary, not real. So both characters are using math terms as insults, and the punchline works on two levels: logic and math.
