Answer

Transport vesicles first form in the transitional portion of the ER. Once released from the ER they travel to the Golgi Apparatus where they fuse with cisternae and various enzymes in the Golgi help in the maturation process for the materials within the vesicle. At this point the vesicle will fuse out of the Golgi and head towards the plasma membrane where it would leave the cell (this would occur if the contents within the vesicle were needed in a different cell) or the vesicle may head to a different location within the same cell or it may be sent back to the ER (this would occur if the contents within the vesicle were needed at the ER).
