Why does Friar Lawrence seem so uneasy about Paris's request? How is this dramatic irony?

The Friar is uneasy about Paris' request because he has already officiated over Juliet and Romeo's wedding. That is the irony.... he's being asked to marry (officiate over the nuptials) a woman who is already married to another man.
