Audiences know that the knight guarding the Holy Grail has drunk from the Grail, thus ensuring himself eternal life, before the characters in the film do.

Dramatic Irony: Elsa

Audiences know that Elsa is working for the Nazis before Indy and his father do.

Verbal Irony: Marcus

Marcus asks “Is there anyone here who speaks English? Or maybe even ancient Greek?.” This is sarcastic as very few - if any - people speak ancient Greek.
