Did Dr. Jekyll know from an early time that he was capable of two personalities?

Yes, he knows from the beginning. That is why we find he's changed his will at the beginning of the story. From the start, the Dr. realizes that his evil personality may completely encompass him. Thus, he takes great care to make sure that Hyde is easliy able to have everything he needs.
