During replication, the leading strand is continuously replicated and the lagging strand is replicated in fragments called Ozakazi fragments joined together by ligase. A mutant, defective ligase will lead to a replicated DNA duplex with a strand of fragmented DNA. Sedimentation with the alkaline environments of a sucrose gradient yields two bands of DNA: one denoting the intact single strand and the other denoting unspliced fragments.
