Curley has a major case of small guy complex. Curley uses position as the bosses son to intimidate the men. Few are actually intimidated by Curley himself rather than he is the boss's son. These men desperately need this job. Curley threatens other workers over trivial matters (like where his wife is) but backs down if someone actually challanges him.

JUst to add, Curley of course goes after Lennie in the scene you mention. Lennie is large and seemingly unable to protect himself,

"What the hell you laughin' at?....Come on, ya big ba---. Get up on your feet. No big son---is gonna laugh at me. I'll show ya who's yella."
