Vladek wants to flee to Hungary because he believes that he and Anja can rebuild their lives there and be happy. The couple is captured when the smugglers desert them on the streetcar, and they are taken by the Gestapo. The letter from Mandelbaum's nephew was significant because it let them know all was well in Hungary.
