Lennox probably began suspecting Macbeth at the end of act 3. He has close relations to Macduff who definitely suspects Macbeth for the killing of Duncan. Lennox also seems to know the latest news in Scotland after Macduff flees. He fills people in on what a tyrant Macbeth has become.
