How has lady Macbeth changed since the beginning of the play?

In Act III, Scene II, Lady Macbeth becomes increasingly anxious. She is no longer the tough woman we met at the start of the play, she is beginning to crumble.... she is filled with insecurity.

Naught’s had, all’s spent,
Where our desire is got without content.
'Tis safer to be that which we destroy
Than by destruction dwell in doubtful joy.
