The man tells the conductor that Buck is being taken to San Francisco in order to cure his "fits." Buck and the man have a series of fights for dominance, and the man thinks that he will be able to stop this instinctual drive in Buck.
I think it is because Buck simply would not be subervient to the men putting a rope around his neck. The doctor feels he can tame him.
Why was Buck treated so poorly by all these men, especially the driver in the red shirt?
